Title The Effect of Cisplatin on the Calcium Regulation in Isolated Outer Hair Cells
Purpose In this study, authors aimed to know the changes of calcium channel regulation by cisplatin in isolated outer hair cells and to speculate the mechanism of hearing loss by cisplatin.
Materials and Methods Outer hair cells were isolated from guinea pig after enzymatic and mechanic dissociation. Calcium current was recorded by whole cell patch clamp method and intracellular calcium was measured by Fura 2-AM fluorescent dye. Cisplatin solution (0.3 mM) was perfused in bath solution and the changes of calcium current and intracellular calcium were recorded.
Results In outer hair cell, calcium current was decreased after cisplatin perfusion, but the shape of current-voltage curve did not change. Intracellular calcium was also decreased after cisplatin perfusion, but after repeated perfusion, intracellular calcium increased even after perfusion with nifedipine.
Conclusion Calcium channel can be blocked and intracellular calcium store may be disturbed by cisplatin. Therefore, repeated exposure of cisplatin can increase intracellular calcium and make the cells dead.
